MASS SPECTROMETRY

Peptide identity

Is it the peptide you expect?

Compare the observed molecular mass with the expected mass of the declared peptide. A mass match supports an identity assessment; it does not, by itself, prove the full amino-acid sequence.

Sequence confirmation or differentiation of isomers requires additional, specifically agreed methods.

HPLC–UV

Chromatographic purity

What does the chromatogram reveal?

Separate the components of a research sample and report the principal peak as a percentage of the integrated UV peak area under defined conditions.

UV area percentage is not the same as peptide mass fraction, peptide content or a comprehensive screen for all impurities.

QUANTITATIVE ANALYSIS

Peptide content

How much peptide is present?

Determine peptide quantity using an agreed quantitative procedure and a suitable reference standard. Results are reported in the units appropriate to your sample.

The method, standard availability, sample requirements and reporting basis are agreed before testing.

ICP–MS

Elemental impurities

Are trace metals present?

Screen for lead (Pb), arsenic (As), cadmium (Cd) and mercury (Hg). Each element is reported individually with its concentration and applicable reporting limits.

Detection capabilities depend on the validated method and sample matrix. ICH Q3D is not a universal concentration limit for research peptides.
